FG 2216 is an erythropoietic small molecule prolyl hydroxylase inhibitor, originally developed by FibroGen, that is undergoing development for the treatment of anemia. PDH inhibitor FG-2216 was orally bioavailable and induced significant and reversible EPO induction in vivo. Chronic oral dosing in male rhesus macaques was well tolerated, significantly increased erythropoiesis, and prevented anemia induced by weekly phlebotomy. Furthermore, modest increases in HbF-containing red cells and reticulocytes were demonstrated by flow cytometry. FG 2216 had been in phase II clinical trials for the treatment of anemia, however, all researchers on this drug candidate were discontinued.
JNJ 42041935 is a potent, 2-oxoglutarate competitive, reversible, and selective inhibitor of PHD enzymes. Administration of JNJ-42041935 to rats for 5 consecutive days resulted in a 2-fold increase in reticulocytes, an increase in hemoglobin, and an increase in the hematocrit. It may be an effective means to treat anemia of various origin, in which current treatments are ineffective or not optimal.

Daprodustat (DUVROQ) is a small molecule inhibitor of hypoxia-inducible factor prolyl hydroxylase (PHD) developed by GlaxoSmithKline for the treatment of anaemia in patients with chronic kidney disease (CKD). Inhibition of PHD prevents degradation of hypoxia-inducible factor (HIF), leading to the production of erythropoietin and subsequent induction of erythropoiesis. In June, daprodustat received its first approval in Japan for the treatment of renal anaemia.

Hypotaurine (2-aminoethane sulinic acid) is the immediate precursor of taurine (2-aminoethane sulfonic acid) which is one of the most abundant free amino acids in the central nervous system. Although hypotaurine and taurine are mainly supplied from blood plasma across the blood-brain barrier in the brain, they are also synthesized from cysteine and cysteamine by astrocytes and neurons. Hypotaurine acts as an endogenous neurotransmitter via action on the glycine receptors. It was shown, that hypotaurine might be used as an effective therapeutics for orofacial pain. Also in vitro studies were conducted to show that hypotaurine activates hypoxia signaling through the competitive inhibition of prolyl hydroxylase domain-2. Moreover, was made a suggestion, that the hypotaurine metabolic pathway may provide a potential new target for glioblastoma diagnosis and therapy.
