Pridinol mesilate is a centrally acting muscle relaxant that is used in the symptomatic treatment of muscle spasm. It is also used as the hydrochloride salt for its antimuscarinic activity in the management of parkinsonism.

Spasmolytic drug pramiverine was used in the active treatment of labor. In addition, it participated in clinical trials for patients with colitis syndrome. It was revealed, that pramiverine had improved the conditions of patients.

Neridronic acid (6-amino-1-idroxyesilidene-1,1-bisphosphonate) is a nitrogen-containing bisphosphonate licensed in Italy for the treatment of osteogenesis imperfecta and Paget’s disease of bone. The pharmacodynamic profile is similar to that of other nitrogen-containing bisphosphonates and is characterized by its high affinity for bone tissue particularly at sites undergoing a process of remodeling. In growing children affected by osteogenesis imperfect, neridronic acid rapidly increases bone mineral density as measured by dual X-ray absorptiometry and this is associated with a significant decrease in fracture cumulative number. Similar results have been obtained also in newborns (<12-month-old) and in adult patients. In Paget’s disease of bone, 200 mg intravenous neridronic acid is associated with a 65% rate of full remission and a biochemical response (decrease of > 75% of bone turnover markers) in 95% of the patients. Neridronic acid treatment has been reported to be effective also in other skeletal diseases such as osteoporosis, algodystrophy, hypercalcemia of malignancy and bone metastasis. Neridronic acid has been developed only for parenteral use, and it is the only one used as the intramuscular injection. This avoids all the limitations of oral bisphosphonates and may be offered for a home treatment with simple nursing assistants

Noxiptiline (brand names Agedal, Elronon, Nogedal), is a tricyclic antidepressant (TCA) that was introduced in Europe in the 1970s for the treatment of depression. In a double-blind comparison of noxiptiline and amitriptyline in hospitalized patients with primary depressive illness for 3-6 weeks there were no significant differences, but noxiptiline had a faster onset of action. A comparison with imipramine also showed no difference in therapeutic efficacy. As regards adverse effects, noxiptiline seemed to cause more mental symptoms, such as delirium. In one study in 44 patients, 9 developed delirium, there were mild manic symptoms in 6, dry mouth in 21, disturbed micturition in 2. and difficulty in accommodation in 3.

Fipexide (aka attentil and vigilor) is a psychoactive drug of the piperazine class which was developed in Italy in 1983. It was used in Italy and France for the treatment of senile dementia but is no longer in common use due to undesirable side effects including fever and hepatitis.

Bucloxic acid, a nonsteroidal anti-inflammatory agent, which was studied to treat the chronic glomerular nephropathy.

Temocillin was marketed by Beecham Pharmaceuticals in the UK in the 1980s but achieved little commercial success and was withdrawn, though it remained available via the manufacturer’s medical department. Presently licensed to Eumedica, temocillin is being re-launched in the UK and Belgium for treating UTI, sepsis, and respiratory infections by ESBL (Extended-spectrum beta-lactamases) and AmpC-producing Enterobacteriaceae. It acts by inhibiting the synthesis of the peptidoglycan layer of bacterial cell walls. It irreversibly binds to the active site of specific transpeptidases and carboxypeptidases known as Penicillin Binding Proteins (PBP), preventing peptidoglycan production.

Tolonidine, 2(2-chloro-p-toluidino)-2-imidazoline-nitrate, is a substance chemically related to clonidine. In the anesthetized dog, tolonidine administered i.v. decreased the amplitude of ventricular contractions, reduced aortic blood flow and increased peripheral vascular resistances. In the bivagotomized pithed rat, tolonidine induced a long-lasting increase in blood pressure with no secondary hypotension, thus suggesting peripheral sympathomimetic properties, however, contractions of seminal vesicles in vitro were not obtained. The product proved to have no peripheral sympatholytic or parasympatholytic properties.

Benexate is an active ingredient of benexate.CD or benexate hydrochloride betadex, a beta-cyclodextrin complex. Benexate was approved for the treatment of Gastric Ulcer in Japan. The terapeuthic effect of benexate is associated with promotion of prostaglandin synthesis, inhibition of acid secretion and increase in mucosal blood flow.
