Emodepside is a semi-synthetic product (originated by Astellas and out-licensed to Bayer for animal and human use); its precursor is synthesized by a fungus living in the leaves of Camellia japonica. It is a potent antihelminthic drug used in combination with praziquantel (as Profender®) and in combination with toltrazuril (as Procox®) for the treatment of parasitic worms in cats and dogs. Emodepside, a semi-synthetic derivative of PF1022A, belongs to a new class of anthelmintic drugs, the cyclooctadepsipeptides, and shows good efficacy against macrocyclic lactone-, levamisole- or benzimidazole-resistant nematode populations. Although putative receptors for emodepside have already been discovered, its mode of action is still not fully understood. It has being suggested that GABA(A)-receptor UNC-49 is associated with the emodepside mode of action. It has also being shown that Emodepside binds to a presynaptic latrophilin receptor in nematodes. The following presynaptic signal transduction occurs via activation of Gqalpha protein and phospholipase-Cbeta, which leads to mobilization of diacylglycerol (DAG). DAG then activates UNC-13 and synaptobrevin, two proteins which play an important role in presynaptic vesicle-functioning. This finally leads to the release of a currently unidentified transmitter. The transmitter (or modulator) exerts its effects at the postsynaptic membrane and induces a flaccid paralysis of the pharynx and the somatic musculature in nematodes.

Imidacloprid is a systemic, chloro-nicotinyl insecticide used for the control of sucking insects such as fleas, aphids, whiteflies, termites, turf insects, soil insects, and some beetles. It is used on co on and vegetable crops as foliar and seed treatments, soil, structures, indoor and outdoor insect control, home gardening and pet products. It is indicated for the prevention of heartworm disease caused by Dirofilaria immitis. It kills adult fleas and is indicated for the treatment of flea infestations (Ctenocephalides felis). It is also indicated for the treatment and control of the following intestinal parasites Hookworm species, Roundworm species, Whipworms. Adverse events in animals included: malaise, vomiting, diarrhea, shaking, mydriasis, hypersalivation with abnormal neurologic signs, seizures, death, generalized hematoma of the body, and alopecia at the treatment site. Adverse reactions in humans included: burning, tingling, numbness, bad taste in the mouth, dizziness, and headache.

Selamectin (trade name Revolution, Stronghold) is a broad-spectrum endectocide. It is used in dogs and cats for treatment, control and prevention against fleas, heartworms, ear mites, ticks and other parasites. Selamectin is not approved for human use. It is a a semi-synthetic compound of the avermectin class, a macrocyclic lactone.
Pyriproxyfen is a broad-spectrum insect growth regulator with insecticidal activity against public health insect pests: houseflies, mosquitoes and cockroaches. In agriculture and horticulture, pyriproxyfen has registered uses for the control of scale, whitefly, bollworm, jassids, aphids and cutworms. Pyriproxyfen is used on citrus fruit in Israel, South Africa, Spain and Italy. Pyriproxyfen is one of several insecticides used for the control of the red imported fire ant (Solenopsis invicta) in California, USA. Pyriproxyfen has also been considered by WHO for vector control under its Pesticides Evaluation Scheme. It is a potent suppressor of embryogenesis and adult formation of the sweetpotato whitefly, Bemisia tabaci (Gennadius), and the greenhouse whitefly, Trialeurodes vaporariorum (Westwood). Dipping of cotton or tomato seedlings infested with 0 to 1-day-old eggs in 0.1 mg litre−1 resulted in over 90% suppression of egg hatch of both B. tabaci and T. vaporariorum. Pyriproxyfen is registered in the U.S. for flea and tick control in the home and on pets, as well as indoor and outdoor ant and roach control. Formulas include carpet powders, foggers, aerosols, shampoos, bait, and pet collars.

Dinotefuran is a new furanicotinyl insecticide which represents the third generation of neonicotinoid group. Dinotefuran, which was developed by Mitsui Chemicals, Inc., was registered in Japan April 24, 2002 and the registration has been submitted to the Environment Protection Agency (EPA) in the USA. Dinotefuran was granted Organophosphorus Alternative and Reduced Risk Status by the EPA. Dinotefuran exhibits a insecticidal activity against Hemiptera, Lepidoptera, Coleoptera, Diptera, Dictyoptera and Thysanoptera. Furthermore, it has very low phytotoxicity so that it can be utilized for many kinds of crops. Dinotefuran is water-soluble and has excellent systemic and translaminar action in many plants. This property enables dinotefuran to be applied using various methods and various formulations. The binding assay using insect nAChRs and the electrophysiological study showed that dinotefuran acted on nAChRs as an agonist. However, in the binding study, the affinity of dinotefuran against the binding site of other neonicotinoids was very low, suggesting that this compound acts on a different site than other neonicotinoids.
Indoxacarb is a new oxadiazine insecticide discovered and developed by DuPont. The indoxacarb racemate contains two enantiomers (S : R), designated DPX-KN128 and DPX-KN127, but only the S enantiomer has insecticidal activity. It inhibits insect neuronal voltage-dependent sodium channels. Indoxacarb is an active component of Merck Activyl® used in the treatment of flea and tick in dogs and cats. DuPont™ Steward® WG is a water dispersible granule stomach and contact insecticide powered by Indoxacarb for control of various insect pests on a variety of crops.

Ibafloxacin is a fluoroquinolone antibiotic drug used in veterinary medicine. Ibafloxacin, a fluorinated 4-quinolone, is a broad spectrum antibiotic with bactericidal action against Gram-positive and Gram-negative bacteria. Ibafloxacin is presented in the form of a tablet/oral gel containing a racemic mixture of S- and R-ibafloxacin. The antimicrobial activity of the racemate originates mainly from the S-enantiomer. Ibafloxacin was marketed under the brand name Ibaflin. Ibaflin tablets were intended for use in dogs for treatment of respiratory tract infections, urinary tract infections and dermal infections caused by ibafloxacin susceptible pathogens. Ibafloxacin, is an antibiotic belonging to the class ‘fluoroquinolones’. It works by blocking an enzyme called ‘DNA gyrase’, which is important in allowing bacteria to make copies of their DNA. This enzyme is only found in bacterial cells, and does not have a similar function in animal cells. By blocking DNA gyrase, ibafloxacin prevents the bacteria from making DNA and stops them making proteins and growing, resulting in their death.
Fipronil is a member of the phenyl pyrazole class of pesticides, which are principally chemicals with a herbicidal effect. Fipronil, however, acts as an insecticide with contact and stomach action. Fipronil is an extremely active molecule and is a potent disruptor of the insect central nervous system via the (-aminobutyric acid (GABA) regulated chloride channel. Fipronil is classed as a WHO Class II moderately hazardous pesticide. Cutaneous skin reactions appear to be the most common adverse drug experiences reported for fipronil in dogs and cats.Animals exposed to large doses of fipronil via accidental oral absorption are expected to display seizures, and neurological and hepatic function should be monitored. There appear to be no reported interactions between fipronil and approved companion animal drugs and ectoparasiticides.

Spinosad is a natural mixture of pediculicidal tetracyclic macrolides—spinosyn A and spinosyn D in the ratio of 5:1. It is derived from species of actinomycetes bacteria - Saccharopolyspora spinosa and is a bacterial waste product produced by fermentation on a nutrient food source. It has since long been used as a pesticide and classified by the US Environment Protection Agency as a reduced risk pesticide product. Spinosad has a high level of efficacy for lepidopteran larvae, as well as some Diptera, Coleoptera, Thysanoptera, and Hymenoptera, but has limited to no activity to other insects and exhibits low toxicity to mammals and other wildlife. Spinosad overstimulates nerve cells by prolonging electrical impulse across synapses by acting like acetylcholine. After periods of hyperexcitation, lice become paralyzed and die. Recently, FDA has approved the topical suspension of spinosad 0.9% for treatment of head lice infestation in patients four years of age and older. It is both pediculicidal and ovicidal.
