Dimethiodal is the diagnostic aid (radiopaque medium). Dimethiodal sodium was the chemical analog of methiodal sodium with two instead of one iodine atom per molecule. It is an uro-angiographic contrast agent that, together with methiodal sodium, iodomethamate disodium and iodopyracet diethanolamine, dominated the market in 1930s and 1940s.

Nicocodine is an opioid related to codeine. It is also an antitussive agent. Nicocodine exhibits at least twice the potency of codeine in clinical experiments. The study of the pharmacokinetic behaviour of nicocodine by means of blood and brain level curves of rats after i .v. application showed that the penetration of the blood
brain barrier seems to be favoured for nicocodine. The detected peak concentration of nicocodine in brain after i.v. application is 4.4 times higher than the blood level values at the same time - codeine the main metabolite is detected in almost equal amounts in brain and blood. A comparative assay of codeine and nicocodine after p.o. application of equimolar doses per kg body weight revealed that predominantly codeine is found in brain and its peak value after nicocodine administration is 3-fold higher than after codeine administration. Nicocodine is hydrolysable to morphine and the WHO Expert Committee on Addiction-Producing Drugs (1962) recommended its international control as a narcotic, like other convertible drugs in the morphine series.

Silperisone is a tolperisone like organosilicon compound with centrally acting muscle relaxant properties. Studies in mice showed that silperisone may have less propensity to cause CNS depressant or motor side effects than tolperisone or other antispastic drugs. In cats and rats, silperisone was an effective suppressant of monosynaptic and polysynaptic spinal reflexes and decerebrate rigidity. Its suppressant effect on the spinal reflexes was also demonstrated in the isolated hemisected rat spinal cord in vitro. The in vivo potency and efficacy of silperisone by i.v administration were similar to those of tolperisone and eperisone. The mechanism underlying the spinal reflex depressant effects of silperisone involves the blockade of voltage gated neuronal sodium and calcium channels leading to a decreased release of excitatory transmitter and reduced neuronal excitability. In addition, silperisone has potassium channel blocking effect. Phase I clinical studies with silperisone at doses up to 150 mg/day failed to detect any adverse effects at plasma concentrations considered to be effective in the preclinical tests. These findings suggested that silperisone might be a useful antispastic drug. However, findings in chronic animal toxicity studies led to the discontinuation of silperisone's development.

Tanaproget (WAY-166989) is a nonsteroidal progesterone receptor agonist. It has been in Phase-II clinical trials as an oral contraceptive. The compound demonstrated a positive preclinical pharmacological profile in the treatment of endometriosis. The level of progesterone receptors in breast tumours can be used to guide the selection of endocrine therapies for breast cancer patients. Radiolabeled analogues of tanaproget have diagnostic potential as PET imaging agents for breast cancer.
