(+)-DDMS (R-Didesmethylsibutramine , (R)-DDMS) is one of sibutramine active metabolites. Sibutramine is widely used in the treatment of obesity. Sibutramine acts by inhibiting the reuptake of serotonin and noradrenaline in synapses, thereby enhancing both satiety and energy expenditure. In preclinical models (R)-enantiomer of Didesmethylsibutramine was clearly more potent than the (S)-enantiomers and (R)-didesmethylsibutramine shows some activity in all tests. (S)-didesmethylsibutramine affected locomotor behavior and the Porsolt test but appeared to be completely inactive on food intake. R-Didesmethylsibutramine is more potent than sibutramine in depressing food intake and decreasing body weight, suggest that these enantioselective metabolites might be safer and more effective than sibutramine as potential therapies for obesity.

Mesaconitine is a diterpene alkaloid, from the plants of the Aconitum genus, Ranunculaceae. Mesaconitine is a centrally acting analgesic without affinity to opioid receptors. It has been reported that the antinociception is due to an interaction with the noradrenergic system. As a neurotoxic, it opens the TTX-sensitive Na+ channels in the heart and other tissues and induces arrhythmia. The activity of Mesaconitine, which is a chemical analog of Aconitine, on TTX sensitive Na+ channels in the heart and other tissues is stronger than that of Aconitine. The duration of its neurotoxic effect is shorter.
It possesses an hypotensive activity. The hypotensive action is not inhibited by an adrenergic b-blocker but abolished with muscarinic blocker. It may be partially mediated by a muscarinic mechanism. Mesaconitine binds with high affinity to the open state of the voltage-sensitive sodium channels at site 2, thereby causing a persistent activation of the sodium channels, which become refractory to excitation.

Du-24565 is a potent and selective inhibitor of the synaptosomal uptake of serotonin. At higher concentration, it affects the uptake of norepinephrine and dopamine. It is a potential antidepressant and can be useful as a pharmacological tool to study the role of 5-HT in the central nervous system. DU-24565 had little effect on serotonin or tyrosine accumulation in human melanoma cells but suppressed the uptake of extracellular Dopa. In vitro, DU-24565 inhibited the proliferation of PC-3, DU-145 and LNCaP human prostate carcinoma cells in a dose-dependent manner.
