Elisidepsin (Irvalec®, PM02734) is a depsipeptide produced by chemical synthesis and chosen for development as an antineoplastic agent based on its in vitro activity against human solid tumor cell lines, in vivo activity in hollow fibers and xenografted human tumors, as well as its acceptable preclinical toxicology profile. Elisidepsin causes a typical necrotic cell death and induces profound alterations in tumor cell morphology. Elisidepsin inserts into the plasma membrane causing rapid loss of integrity and necrotic cell death in tumor cells. PharmaMar was developing elisidepsin for the treatment of cancer. However, development of the compound has been suspended.

Ozarelix is a luteinizing hormone-releasing hormone (LHRH) antagonist. It is known that LHRH antagonist exerts rapid inhibition of luteinizing hormone and follicle stimulating hormone with an accompanying rapid decrease in sex hormones. Thus this inhibitor can be effective in a variety of hormonally dependent disease including prostate cancer, benign prostatic hyperplasia (BPH), and endometriosis. Ozarelix was developed for the treatment of all these diseases including Alzheimer's disease. However, in January 2010 Spectrum Pharmaceuticals announced that it was discontinuing development of ozarelix in BPH. Because the low-dose intermittent therapy was disappointing in the treatment of lower urinary tract symptoms in men with BPH. The development of the drug for Alzheimer's disease was also discontinued. Ozarelix completed phase II trials for the treatment of prostate cancer and is in preclinical trials for the treatment of endometriosis and ovarian cancer.

UM171 is a potent agonist of human hematopoietic stem cell renewal, independently of AhR suppression. UM171 act differently than other small molecule stimulators of hematopoiesis, such as the aryl hydrocarbon receptor (AhR) antagonist StemRegenin 1 (SR1). Addition of UM171 to cultures containing SR1 and cytokines further enhances the ex vivo expansion of normal HSCs, including CD34+ cells.

Evenamide (NW-3509) is a blocker of voltage-gated sodium channels. Evenamide modulates sustained repetitive firing, without inducing impairment of normal neuronal excitability. It normalizes glutamate release induced by aberrant sodium channel activity. The potential benefits of the compound have been demonstrated in numerous preclinical models predictive of efficacy in psychiatric diseases, including models of psychosis such as amphetamine-induced hyperactivity, sensorimotor gating and information processing deficits (pre-pulse inhibition impairment induced by different stimuli), mania and depression. Evenamide is being evaluated in a Phase II trial as add-on treatment to 5HT2/D2 blocking antipsychotics in schizophrenic patients.
