MFZ 10-7 is a high affinity negative allosteric modulator at the mGlu5 receptor (Ki = 0.67 nM). MFZ 10-7 demonstrates anxyolitic properties in mouse models. Intraperitoneal administration of MFZ 10-7 inhibited intravenous cocaine self-administration, cocaine-induced reinstatement of drug-seeking behavior and cocaine-associated cue-induced cocaine-seeking behavior in rats.

PD 144418 oxalate is a remarkably potent and selective sigma1 receptor ligand having potential antipsychotic properties. PD 144418 oxalate displayed an apparent affinity (Ki) of 0.08 nM for sigma1 sites in guinea pig brain membranes and a Ki of 1377 nM for sigma2 sites in rodent neuronal NG108-15 cell membranes. PD 144418 oxalate was classified as sigma1 receptor antagonist based upon several assays, including the ability to attenuate mescaline-induced scratching in mice. PD 144418 oxalate attenuates cocaine-induced hyperactivity in mice. It showed potential antipsychotic activity, but no antidepressant or anxiolytic actions.
77-LH-28-1 Oxalate is a salt of 77-LH-28-1. This drug is a potent, selective, bioavailable and brain-penetrant agonist of muscarinic acetylcholine receptor subtype 1. 77-LH-28-1 can increase network oscillations relevant to cognitive processing that would support the potential use of this mechanism in the treatment of cognitive impairment. 77-LH-28-1 exhibited U-shaped dose-effect functions in attenuating cocaine discrimination. 77-LH-28-1, can significantly enhance donepezil-induced gamma oscillations. These data support the notion that it should be possible to find a more efficacious AChE inhibitor or an adjunctive approach, to provide a better therapeutic intervention in Alzheimer's disease.
