HM-30181 is a highly selective and potent inhibitor of Multi-drug resistance 1 (MDR1, ABCB1), also known as P-glycoprotein (P-gp). Co-administration of HM30181 greatly increased oral bioavailability of tubulin-stabilizing chemotherapeutic agent paclitaxel. Oraxol is an oral dosage form of paclitaxel administered orally with the HM30181A molecule. Oraxol offers patients with paclitaxel-responsive tumors the possibility of oral therapy without the requirement for premedication to prevent infusion-related hypersensitivity-type reactions. Current clinical data suggests the promising potential of a better clinical response and tolerability profile, which can likely to be attributed to the better pharmacokinetic profile achieved. Oraxol is presently in a Phase 3 trial in metastatic breast cancer and poised to enter into a combination study for treatment of advanced gastric cancer with ramucirumab through a clinical trial collaboration with Eli Lilly and Company.

Centanafadine is an oral triple reuptake inhibitor that has been developed by DOV Pharmaceutical as a treatment for attention-deficit hyperactivity disorder (ADHD). Centanafadine works by modulating the activity of norepinephrine, dopamine, and serotonin, three neurotransmitters known to be relevant in patients with ADHD. In the human abuse liability study, immediate-release centanafadine demonstrated a reduced abuse potential compared with the schedule II stimulants d-amphetamine and lisdexamfetamine. Treatment with high doses of immediate-release centanafadine resulted in a markedly different profile than that of the comparators, with most subjects experiencing the acute onset of adverse effects, including nausea, vomiting, and dysphoria. Almost 2 hours after the administration of centanafadine, the test subjects reported “liking” at about two-thirds of the magnitude of amphetamines, a finding that may have indicated dopamine activity. However, unlike amphetamines, which provided an immediate positive experience, the subjects receiving centanafadine experienced negative effects before reaching that point.

KYORIN Pharmaceutical has developed a new sphingosine 1-phosphate (S1P) receptor type 1 agonist, 2-amino-2-propanediol hydrochloride (KRP-203), for immunomodulation in autoimmune diseases and organ transplantation. This drug was in phase II clinical trial for the treatment Ulcerative Colitis, but this study was terminated. In addition, it has been investigated for the treatment of Cutaneous Lupus Erythematosus and Crohn's Disease, these phases II clinical trial studies were successfully completed.

In animal models of pancreatic neuroendocrine tumors, this compound prevents lymph node metastasis. PF-04217903 had been in phase I clinical trials by Pfizer for the treatment of solid tumors. In 2011, the company discontinued the development of the compound.

Alagebrium (formerly known as ALT-711) - is one of the better known compounds discovered and developed with the intent of repairing a type of accumulated damage that leads to age-related degeneration. In this case, the damage is the buildup of extracellular protein crosslinks, and amongst them the type known as advanced glycation endproducts. The formation of advanced glycation end-products is associated with arterial stiffness in experimental models and alagebrium, an advanced glycation end-product cross-link breaker, has been shown to reduce arterial stiffness in elderly subjects. Alagebrium is an AGE-lowering agent with beneficial effects in renal structural and functional parameters in diabetes, decreased diabetes-accelerated atherosclerosis, and age-related myocardial stiffening. ALT-711 exhibits a structural homology to thiamine, and it was suggested to interfere with thiamine metabolism. Enzyme kinetic experiments showed that ALT-711 dose-dependently decreased TDPK activity with K(i)s, calculated by different experiments and fitting models ranging from 0.88 to 1.09 uM. Alagebrium has been investigated for the treatment and prevention of aging, heart failure, physical activity, diabetic nephropathy, and cardiovascular disease, among others. However, this research has been discontinued.

Voruciclib (also known as P1446A-05) is a flavone-based, potent and selective CDK 4/6 inhibitor with activity in multiple BRAF-mutant and wild type cell lines. It is currently in clinical trials in combination with BRAF inhibitor (PLX4032) to treat advanced BRAF-mutant melanoma. Voruciclib has significant inhibitory activity against cutaneous and uveal melanoma. Mechanistic studies revealed that P1446A-05 inhibits phosphorylation targets of CDK members, and induces cell cycle arrest and apoptosis irrespective of melanoma genotype or phenotype. Voruciclib Hydrochloride is in phase I clinical trials by Piramal Life Sciences for the treatment of chronic lymphocytic leukaemia and malignant melanoma.

Phenadoxone hydrochloride is one of some forty amino-ketones and amino-esters related to amidone. The compound is a very potent analgesic for the rat; by the subcutaneous route it is more active than either morphine or amidone. In spite of this its acute toxicity to mice is lower than that of amidone and its therapeutic index is therefore correspondingly higher, giving a wider margin of safety. Side effects in dogs, such as narcosis, sedation, and general depression, were much less with phenadoxone than with amidone or morphine. Nausea and vomiting did not occur after phenadoxone in non-tolerant dogs. Clinical results show that for relieving certain types of pain in human subjects it is a potent analgesic that compares favorably with morphine and amidone. At therapeutic dose levels undesirable pharmacological effects, such as cardiac depression and vasomotor disturbance, are absent, and it is only at extremely high dose levels that untoward effects occur. However, the drug has a strong respiratory depressant action when given in high doses; it should be used with special caution if injected intravenously.

Nutlins are cis-imidazoline analogs which inhibit the interaction between mdm2 and tumour suppressor p53. Inhibiting the interaction between mdm2 and p53 stabilizes p53, and is thought to selectively induce a growth-inhibiting state called senescence in cancer cells. These compounds are therefore thought to work best on tumors that contain normal or "wild-type" p53. Nutlin-3 does not induce the phosphorylation of p53 on key serine residues and reveals no difference in their sequence-specific DNA binding and ability to transactivate p53 target genes compared with phosphorylated p53 induced by the genotoxic drugs doxorubicin and etoposide, demonstrating that phosphorylation of p53 on key serines is dispensable for transcriptional activation and apoptosis.
