Thiophanate methyl is an approved EU fungicide used widely to control fungal diseases on crops. It has a low aqueous solubility, low volatility and tends not to be persistent in soil or water systems. It has a low mammalian toxicity, however it is an irritant, a skin sensitiser and may also be mutagen. It is moderately toxic to most aquatic organisms and earthworms but less so to birds and honeybees.

Following the oral administration of 700 and 1000 mg kg(-1) body wt. for five consecutive days, short-term testicular toxicity was examined in the B6C3F1 mouse through specific parameters (sperm head count, specific enzyme activities, histopathology on days 3-35 post-dosing).

Curator's Comment: In the absence of metabolic activation, Thiophanate methyl exerted cytotoxic activity in BALB/c 3T3 cells, evidenced through the formation of cell colonies, at low doses (> 10 ug/ml). In the presence of metabolic activation, the cell-transforming activity was detectable at all tested doses (i.e. from 20 to 200 ug/ml) and it was particularly evident in a level-II transformation amplification test when the cells were allowed to perform active proliferative activity. https://www.ncbi.nlm.nih.gov/pubmed/9434840
Interaction of a genotoxic fungicide Thiophanate methyl (MT) has been studied in vitro with calf thymus DNA. Fluorescence quenching data revealed the binding constant (K(a)=3.23 x 10(4)M(-1)) and binding capacity (n=1.1) of MT with ctDNA.
