ATB-346, being developed by Antibe Therapeutics, a Toronto-based pharmaceutical company, is a hydrogen sulfide-releasing non-steroidal anti-inflammatory drug (NSAID) that inhibits COX and suppresses prostaglandin production. ATB-346 exhibits anti-inflammatory, analgesic, antinociceptive, immunomodulatory, and neuroprotective activities. In vivo, this compound attenuates zymosan-induced inflammation, nociception, and immune signaling. ATB-346 also prevents ligature-induced periodontal bone loss and pathologies, potentially by suppressing increases in pro-inflammatory cytokine levels. Additionally, ATB-346 decreases edema and improves neurological function in animal models of traumatic brain injury (TBI). ATB-346 completed Phase 1 clinical studies in Q1 2015. To better understand the metabolism of ATB-346, Antibe conducted a radiolabeled study in rats at Covance Laboratories that was completed in Q4 2015. Antibe received approval from Health Canada in March 2016 to conduct a Phase 2 trial of ATB-346 in patients with osteoarthritis of the knee. Preclinical studies of ATB-346 for the treatment intestinal cancer; malignant melanoma; periodontal disorders are in progress.

MPC-3100 is a fully synthetic, orally bioavailable, Hsp90 inhibitor developed by Myriad Pharmaceuticals, Inc for cancer treatment. MPC-3100 targets the N-terminal ATP-binding site of Hsp90 and blocks the activity of ATPase. MPC-3100 shows a broad spectrum anti-proliferative activity against various cancer cell lines, such as HCT-116, NCI-N87 and DU-145. MPC-3100 also inhibits tumor growth in the NCI-N87 gastric cancer xenograft mode. Moreover, pharmacokinetics studies show that MPC-3100 displays a superior oral pharmacokinetics profile, good overall exposure and a reasonable hepatic clearance rate. Phase I clinical studies demonstrate MPC-3100 is safe and tolerated when administered at doses below 600 mg per day

Sepetoprost (also known as ONO-9054), prostaglandin analog was developed as a dual agonist of prostaglandin E3 (EP3) and prostaglandin F (FP) receptors. It is known, that the use of a dual prostaglandin EP3 and prostaglandin F receptor agonists is a novel approach for the reduction of intraocular pressure (IOP) in open-angle glaucoma and ocular hypertension. Sepetoprost was successfully studied in clinical trials phase II for the treatment of open-angle glaucoma, ocular hypertension, and mild open angle-glaucoma.

Pexmetinib (ARRY-614) is a potent, orally bioavailable, dual p38 MAPK/Tie-2 inhibitor with potential antineoplastic, anti-inflammatory and antiangiogenic activities. Pexmetinib inhibited leukemic proliferation, prevented activation of downstream effector kinases and abrogated the effects of TNF-alpha on healthy hematopoietic stem cells. In ex vivo stimulated human whole blood, LPS-induced cytokines was inhibited by Pexmetinib with an IC50 value ranging from 50-120 nM. ARRY-614 inhibited the release of IL-6 from SEA- or LPS-challenged mice with an ED50 value less than 10 mg/kg. Combining Pexmetinib with lenalidomide inhibited both pro-inflammatory cytokines and tumor growth in vivo with higher potency, compared with either agent alone. In dose escalation or expansion cohorts, treatment with Pexmetinib either once daily or twice daily was applied to forty-five patients. Pexmetinib reduced the levels of circulating biomarkers and the p38 MAPK activation of bone marrow.
