Conditions:

Thiazolyl Blue is a membrane-permeable yellow dye that is reduced by mitochondrial reductases in living cells to form the dark blue product. The net positive charge on Thiazolyl Blue appears to be the predominant factor involved in their cellular uptake via the plasma membrane potential. Thiazolyl Blue is used as a direct indicator of cytotoxicity (such as for screening cancer drugs), proliferation and apoptosis. Thiazolyl Blue is also an electron acceptor used for studying NADP-dependent dehydrogenases. Thiazolyl Blue reduction is associated not only with mitochondria, but also with the cytoplasm and with nonmitochondrial membranes including the endosome/lysosome compartment and the plasma membrane. Biological Applications: cell viability assay; microbial growth assays; DNA quantification assays; tissue viability assays; detecting enzymes; measuring membrane potential; treating Alzheimer’s disease, asthma, cancer. Because Thiazolyl Blue forms an insoluble formazan it has usually been applied in endpoint assays.

Emoxypine (ethylmethylhydroxypyridine) succinate (MEXIDOL®) is a 3-hydroxypyridine derivative which is quite similar in structure to Vitamin B6 (pyridoxine). The most important components of the mechanism of action of Mexidol® are its antioxidant and membranotropic effects, the ability to modulate functioning of receptors and membrane-bound enzymes, restore a neurotransmitter balance. Due to its mechanism of action Mexidol® has a wide range of pharmacological effects, realized on at least two levels – neuronal and vascular. It has antihypoxic, anti-ischemic, neuroprotective, nootropic, vegetotrophic, anti-stress, anxiolytic, anticonvulsant, anti-alcohol, cardioprotective, anti-atherogenic, geroprotective and other actions. Under the action of Mexidol®, it is observed an improvement in cerebral circulation and microcirculation. Emoxypine is distributed in Russia but it is widely unknown in other regions.

Mobocertinib (EXKIVITY™) is a first-in-class EGFR tyrosine kinase inhibitor being developed for the treatment of EGFR exon 20 insertion (EGFRex20ins) -positive non-small cell lung cancer (NSCLC). Mobocertinib is a kinase inhibitor of the epidermal growth factor receptor (EGFR) that irreversibly binds to and inhibits EGFR exon 20 insertion mutations at lower concentrations than wild type (WT) EGFR. Two pharmacologically-active metabolites (AP32960 and AP32914) with similar inhibitory profiles to mobocertinib have been identified in the plasma after oral administration of mobocertinib. In vitro, mobocertinib also inhibited the activity of other EGFR family members (HER2 and HER4) and one additional kinase (BLK) at clinically relevant concentrations (IC50 values <2 nM). Based on efficacy in patients whose disease had progressed on or after platinum-based therapy in a phase I/II trial, mobocertinib was recently granted accelerated approval in the USA in this indication. The drug is also being assessed for marketing approval in various other countries and territories including the EU and China.
