Hypocrellin B (HB), a natural perylenequinone pigment, has been successfully employed in the photodynamic therapy (PDT) in a variety of human cancer cells due to its high singlet oxygen yield. Hypocrellin B exhibits antibacterial, anti-metastatic, and anticancer chemotherapeutic activities. Hypocrellin B inhibits proliferation of Staphylococcus by increasing ROS levels and damaging cell walls. In ovarian cancer cells, this compound induces apoptosis and inhibits cell migration and adhesion under photodynamic stimulation; it also induces tumor regression in animal models of lymphoma. In vitro, hypocrellin B induces DNA strand breakage.

The mice were randomly divided into 4 groups (n = 5 each) and injected twice (on days 0 and 7) with PBS, free Hypocrellin B (HB), HB-NPs, or HB-P-NPs via the tail vein
(2 mg/kg as HB). 24 h after injection, tumor sites were irradiated with a PDT laser (630 nm, 400 mW/cm2) for 500 sec (200 J/cm2) twice (on days 1 and 8).

Human ovarian cancer HO-8910 cell as a cancer model cell was incubated with hypocrellin B at a concentration of 2.5 uM for 5 h and irradiated by light from a light-emitting diodes (LED) source. Flow cytometry with annexin V/PI staining showed that early apoptotic and late apoptotic (necrotic) rates following photodynamic therapy with hypocrellin B markedly increased to 16.40% and 24.67%, respectively.
