Activation is achieved through tumor microenvironment (TME)-mediated proteolytic cleavage of an internal peptide linker, releasing an inactivating moiety that functionally restricts the molecule’s ability to stimulate T-cells through specific binding to the epsilon subunit of cluster of differentiation 3 (CD3ε; a member of the T-cell receptor complex).
