Results: PET images and TACs showed regional heterogeneity, reflective of the known distribution of PDE10A. Kinetic modelling showed the superiority of the 2TC over the 1TC model (assessed visually and via AIC). SRTM also produced acceptable model fits. 2TC fits produced well-determined VT estimates (COV<10%) for 89/91 TACs. SRTM fits produced well-determined BPND estimates (COV<10%) for 85/91 TACs (remaining 6/91 all SN, all COV<20%). Time stability analysis supported a reduction in scan duration to 60 min. Inter-subject variability was good, with mean COV (n=7) for baseline scans of VT2TC: 21%, BPND2TC: 21%, BPNDSRTM: 18%. Test-retest variability (n=4) was VT2TC: 12%, BPND2TC: 15%, BPNDSRTM: 12%. There was an excellent correlation between the 2TC- and SRTM-derived BPND (r2>0.99 p<<0.05).
Conclusions: Kinetic parameters for (11C)-IMA107 are well-estimated using 2TC and SRTM. Further studies will fully validate the use of cerebellum as reference region.
